cloning vector and restriction mapping - (Jan/05/2009 )

dear all

i have two questions and need your help
1- how can i select the suitable vector for the cloning experiment?
2- in constructing the restriction map. how can i chose the restriction enzymes that i will use in cutting the DNA or there is no basis and i have to try?

Hi

It depends on what restriction sites you need and the selection marker. For instance you could choose pNEB193 for Nde I site with carbenicillin resistance and blue white selection.

Choose the restriction sites on the basis of what you have in the lab and the time the enzyme takes to digest and also its efficiency.

If youare doing it just for experimental purposes, pick any vector for which your lab has a plasmid map. The map will tell you which enzyme sites to expect. For example, pblue-script, or pbr322 or pgem. So many to select from!!
