If you have the primers why not just PCR off the vector. Or if you want to isolate your virus seq then digest with EcoRI, then gel purify the insert from the vector.

I have a Human papillomavirus which cloned in pUC19 in EcoR1 site. I want to extract the HPV DNA and do the PCR for Southern Blot. How can I do this? Is it just use the ECoR1 to cleave it then do the PCR again? thanks for help!
