Baseline and threshold values for a reference and target gene can be different? - (Dec/04/2008 )

Hi,

I have three genes and I will use the ΔΔCT method. I am just wondering do I have to use exactly same baseline (e.g. 3 and 12) and threshold value (e.g. 0.2) for all experiments? Can I calculate the ΔΔCT values having different baseline and threshold values for the reference and target genes?

basically, you can adjust different tresholds and baselines for every target gene including your references.
