i am need of one suggestion from you
i want to clone a plant based gene ( sorce is a plant leaf)
for that i have to make c dna for cloning
please tell me best methods for isolating toal RNA or mRNA from tissue
for that please suggest best methods and kits available in market

Hi, I have used Nucleospin RNA II for izolation of the viral RNA and it works well for me. It is for whole RNA izolation, and I have used reverse transcription with random hex and oligoT primer as well. other possibility is qiagen they provide kits for RNA as well, and I used redzol trizol(phenol based) as well with good purity and concentration as well

for plant extraction of RNA you could use the Qiagen kits, or any kits that have a protocol for plant cell wall digestion. You an also use Trizol, but you have to homogenize it with glass beads, etc.
