sds-gel problem - (Nov/24/2008 )

after running sds-gel, i found a clear band in the gel but at the wrong position, my protein is 37kDa, but the band is at 31kDa, is there anyone that can give me any idea of this? it is the first time i got my protein, i already check the frame is right, and does it ever happen to others that your protein run faster than it suppolsed to be?

Some proteins do not migrate exactly where they are supposed to. If you are absolutely sure that your ab is good, then your protein migrates faster than it normally would!

it must be the same protein. the markers sometimes run faster or slower than your protein sample. some times when I load my marker on the first and last wells this happens. and 31 and 37 are not very different after all. it is sometimes difficult to figure out what the exact size is even when you use markers.

once I also had this problem that NCBI data showd the protein of my interest should have been 45 KDa, but what I got on the gel was 30 KDa.
