How long a membrane can be saved stripping? - (Nov/21/2008 )

Dear all,

Just wonder how long can a membrane be saved for stripping and reprobing without any obvious changes, as I had to leave my membrane, with ECL added, alone for this weekend not looking after.

I have stripping buffer and I've never used it to be honest, but I posted the same question here some time ago and people said we should not let it dry. just take a picture of your membrane and immediately strip and re-do western with another primary.

I have allways stripped my membranes immediately after the first probing, but my lab partner use to wash the membrane with water after ECL, let it dry and keep it at 4 degrees for several days before the stripping... and it works!

I often leave my PVDF membranes on film over the weekend if I don't have time to wait for longer exposures. They generally dry out, but I just re-wet with MeOH, rinse with 1x PBS, and add primary (or if necessary - strip, block, primary). I have never noticed problems with the membranes.
