can you transform directly into liquid culture? - (Nov/19/2008 )

I just want to make some more of a supercoiled plasmid (pBluescript), so I was wondering if I could just transform some DH5alphas and inoculate that directly into an overnight liquid culture with carbenicillin to plasmid prep the next day. I wouldn't do this with ligations, of course, but would there be any problems doing this "shortcut" with a supercoiled plasmid? Presumably the antibiotic selects for non-transformants?

I am pretty sure that is fine. All of your transformants should be the same and the antibiotic will kill all non-transformants.

Besides, if you're just using the culture for plasmid prep, non-transformants that happen to survive will be irrelevant...
