Hi. I was just wondering, if we can re-boil samples already boiled in sample buffer once before? For example if i still have some sample and would like to re-run the gel.

thanks

hi, i try to answer ur question.
in my case, it doesnt matter to keep my sample after boil it in sample buffer and use it again without re-boiling.
but it also may depend on what ur next step.

It makes no sense to "re-boil" samples because they're already denatured and reduced and stay that way for quite long time. If you store your samples for months, you may add some reducing agent and boil again.

thanks alot.... yeap that makes sense, was also wondering what happens if you boil the samples for too long, I am guessing the protein is degraded and you get a streak on the gel?

aggregated rather than degraded (beside you have proteases in your sample)
