Laemmli buffer recipe - (Nov/18/2008 )
I tried to get the recipe for Laemmli buffer online and came with different results. So, which one is correct (OR, could it varies depending on your protein of interest in western?)?
? 25% 2-mercaptoethanol
300 mM 1M Tris-HCl pH6.8
? 0.5% Bromphenol blue
Go to the original paper, it will tell you exactly what they used.
just be careful while weighing bpb as you need a very low amount. don't add too much or it will turn very dark.
Thanks! I also heard of updated versions too. So that's why I could not decide which one to follow. I guess the amount of blue dye can be somewhat flexible, as long as it's not too faint or too dark, but I'm not sure about the amount of mercaptoethanol.
