How to know the genome size? - (Nov/04/2008 )

I am doing a genomic library but I don't know how big is my genome size.
Can anyone tell me how to get the genome size from certain plant species which are not much literature review about that particular plant.
Beside, any recommended protocol to get a pure DNA from fruit pulp ?
Thanks.

Have you looked in PubMed (http://www.ncbi.nlm.nih.gov/sites/entrez)? How about the NCBI Genome site (http://www.ncbi.nlm.nih.gov/sites/entrez?db=Genome&itool=toolbar)? Does Google even have any suggestions (http://google.com)?
Sorry if these questions seem too basic or even insulting to your intelligence. It's just that you have not given us any information as to what reserch you have done so far, so we can only go back to Square One.

sorry for lacking of information.
My question is how to determine the total size of DNA?
I have extracted DNA from dragon fruits pulp and doing project on it. The problem is I didn't know what is the total DNA size.
Even from NCBI and Genebank, I couldn't get any information. Please give me some suggestion.
My friend also doing project on the other fruit but he got the total genomic size from journal.
Just a simple question, is it a must for us to know the total genomic size before we start the library work?
Thank you and sorry for the confusion.

There are a few papers that used flow cytometry to estimate genome size of some plants, eg http://aob.oxfordjournals.org/cgi/reprint/95/1/99
http://crop.scijournals.org/cgi/reprint/45/6/2557
http://aem.asm.org/cgi/reprint/63/10/4001.pdf (melting temperature analysis and DNA reassociation analysis)
