Measuring Ethidium Bromide without DNA - (Nov/04/2008 )

Im doing a project regarding phytoremediation of EtBr on selected grasses.
Im plan to use FT-IR and UV spectrophotometer.
I need some help on my project.

Ques1: I wonder if anyone know what wavelength to detect EtBr alone without any DNA?

The EtBr will be planted in a plastic bag and soil with grass.
Ques 2: Anyone got suggestion how to extract the EtBr from the soil or plant to measure it the EtBr% uptake by the plant.

Sorry I cannot help you with your problem, but when you add EtBr to soil (just curious):

Doesn't the EtBr get bond to (free) DNA in soil????? And are there no interferences with humic acids that have similar structures like DNA???

I had consult my superwiser on that but didn't get the ans. dry.gif
In matter in fact i do think EtBr will intercalate with few DNA in soil and perhaps affect the actual value in my study.
Bout the inteference of humic acid im not so sure bout that. But thanks for bring it up. I will look into it. biggrin.gif
