What grade of primers for real-time TaqMan PCR assays? - (Nov/02/2008 )


Typically the taqman probe in a real-time assay requires HPLC grade. But what about the primers in the assay? Is there a performance difference between using HPLC grade and desalted primers?

I wouldn't say it makes much difference. We always used desalted-only primers for both real-time applications (SYBR, Taqman) and have no problems with that, in Taqman anyway the main specificity lies on the probe, not the primers. HPLC cleanup makes a huge difference in the price of the primers, yet. Probe alone is expensive enough that it doesn't matter.
