gDNA isolation Question - must use tails? (Oct/23/2008 )

Hi all,

I have a quick question. Must I use tail snips (from mouse, if it matters) for isolating DNA for genotyping? I wouldn't think so. If not can I use any tissue/organ (genotyping is the last step in this experiment and I'll no longer have any use for these mice) and can I still use the same isolation method/reagents (proteinaseK, isopropanol, ethanol, etc.) for other tissue types?

Yes! My collegue do this! He cuts a piece of tail, about 3-4 mm, and he obtains a lot of DNA. Well, there's a kit... I don't remember... tomorrow I can tell you the name of the kit.

you can also use a toe if you want.

You can use whatever tissue you want, but it is just plain easier to use a tail cut then other parts for gDNA extraction.
For RNA I would take it from to tissue of interest, but gDNA in the tail is the same as in the tow or ear or spleen.
Just dont use to much tissue (if you use a kit it will be indicated how much)
