a question in protein dialysis - protein dialysis (Oct/11/2008 )

hello everybody:
I have a question in protein dialysis. the first time I dialysis a protein using the pepes system, but considered that the dialysised protein will be used to make protein transfection. so I made the dialysis again but using the PBS system,the problam is that there is a great lost of the protein when I use the PBS system compared with the pepes system. can anybody give me explaination. thank you

It could be aggregation, which should be reversible if you do the dialysis again vs the first buffer (HEPES, maybe??). The protein also could be denatured, in that case, you have to avoid to use PBS.
