How to synthesize your own oligo-dT primer - (Oct/10/2008 )

Hi!

It might sound silly question but how can I synthesize an oligo-dT20 primer for first strand cDNA synthesis without buying it?
Because the commercial ones are expensive...
What the sequence should be?

That's simple, you synthesize a 20 T primer.
You can add some overhang to it, I recommend looking at several product sites how they make it.
I ordered one, because of economy, and it worked just fine.

Yes! For example T(20)VN, where V is anything but T and N is any nucleotide.

We order two oligos, one 15mer (T)15, and another 18mer (T)18. The oligos are just Ts, no overhangs of any kind. When the oligos arrive, just mix them in equal amount to make oligo (dT)15-18 primers with a final concentration of 0.5 ug/ul. It works equally well as oligo(dT) primers we buy from companies. The cost is only a fraction of that.
