Phenol:chloroform:isoamyl pH 5 for DNA extraction? - Kinda worried, only have pH 8 but that should be fine right? (Oct/09/2008 )

ola!

Im trying out a protocol for Purification of my double digested vector and it tells me to use phenol:chloroform:isoamyl pH 5. Now im a bit confused since I thought that one used pH 8 for DNA.

I only found a pH8 solution in the lab and I would really like to try this out today.

Does anyone know why they would write pH 5? Ive asked around in lab and no one knows................

pH5 is for RNA extraction~~
please do not use the ph5 stuff, ur DNA yield will drop dramatically as DNA is less soluble at lower pH i have yet to figure out why . Anybody got any idea and reference??? i m suspecting something to do with its deoxyribose structure !!
