gene dowregulation by miRNA - (Sep/26/2008 )

Hi,

I'm new in the miRNA field, and hoping someone can help me...
I want to do a Luciferase Reporter Assay to see if a specific miRNA is able to downregulate a specific gene. Is it possible to transfect a cell line directly with a miRNA, or do I have to express it on a plasmid?

Actually, you can do both!
It's up to u to decide.
There are commercial ready-to-use miRNAs that you transfect as double stranded mature miRNAs available from many distributors. they're quite expensive if you think that, given time, you can create your plasmids to express whatever miRNA you want.
it depends on how many miRNAs you want to express and on your funds and time! biggrin.gif
