Protein expression using tap water? - (Sep/16/2008 )

our milli-q system is foul and won't be repaired until next week. i need to express proteins *this* week. So has anybody successfully expressed proteins in bacteria using tap water or regular d.i. water? (i will use BL21De3 pLysS cells and pet17b vector; and 2xyt media)

umm... no but I have manage to run (by accident) a PCR using tap water.

Assuming the water is autoclaved, the tap water should be okay. All tap water is doing is adding a few extra ions. (metal ions- which is good for E.coli, carbonate (does not matter). The only problem may be the chlorine, but boiling/autoclaving the water before use will get rid of it.

But if you are worried, could you not try getting MiliQ water from another lab?

thanks for your thoughts! about the other labs, actually the whole building/town is having water issues (too much Fe in the water - nobody knows why. *rolls eyes*). so most likely their milliQ systems are not happy either. oh well --- let's see how it works. i'll probably use D.I. water or steal from somebody else's water stock. thanks again!
