How to design PCR primer to amplify a very long sequence(about 9.7kb) - I want to amplify cDNA genome of HCV. It's about 9.7kb long (Aug/31/2008 )

I want to amplify cDNA genome of HCV. It's about 9.7kb long. I don't know how to design the primer for this sequence.

design the primers just like you would do for any normal pcr..but use some high fidelity pcr mix optimized for long pcr. My pick would be LA TAQ from takara

Search for the term "spilce-overlap PCR". It's a way of amplifying your large genes as pieces and then joining them together. Quite helpful for large genes.
