how to check concentration of dNTP I bought - (Aug/28/2008 )

Recently, I bought a batch of dNTP, but....I suddenly think that the concentration of dNTP in tube are all the same as the concentration on the labe? blink.gif
for example, i bought 100mM, how can i cauclate or check that it is really full of 100mM?

please every frineds give me an answear or methods~~~ wub.gif

Of each dNTP or overall concentration? If you want to distinguish each dNTP you will probably need to do some sort of mass-spectrometry, otherwise there will be a variety of chemical detection methods about.

However, why is it an issue... if you have doubts about the batch, complain to the company and get them to replace the tube, or if you can't be bothered just get a new lot in.

yes, i can request a new one replacement, but still face the same question, how us to believe the concentration is the true.

Maybe the PCR mistake happened on the wrong dNTP concentration but just we too believe the dNTP concentration is sufficient. We never question the dNTP concentration and just check the activity of taq or primer or template. angry.gif

I would be much more likely to question another part of the reaction than the concentration of the dNTPs. If you really doubt the [dNTP], do a PCR that you know works with differing amounts of dNTP. Check your primers, the water you use, and the polymerase, as you said. The [dNTP] is probably one of t he more flexible components in the reaction.
