Silver Stain 2D gels - Trouble with silver staining 2D gels (Aug/26/2008 )

Has anyone had problems with silver staining 2D gels? I have followed standard protocol for running the gels including DTT and iodoacetamide treating the proteins. I first coommassie stain the gels and then try to silver stain. Although the silver stain reagents appear to react, the stain does not seem to penetrate the gels. Any suggestions? Thanks.

normally, reagents are fully water soluble, and should be solved in PAA gels; maybe the CBB stain disturb the binding of Ag; try silver staining w/o prior CBB staining
