chromatin DNA and restriction endonucleases - (Aug/25/2008 )

Hi I am wondering if chromatin DNA (including histone binding with DNA) is able to be digested with restriction endonucleases succesfully? I have tried several times, result is not satisfied. Can you help me?. Thanks a lot.

yes it can be digested (see an example here) but I guess it depends where on the chromatin your restriction site is located (on the spacer DNA, on the histone...)
