siRNA toxicity in MCF7 cells - why are they sick????? (Aug/25/2008 )

Hi!
I'm doing knockdown with siRNA in MCF7 cells and all 4 constructs + scramble make my cells really sick. (25 nM siRNA)
I use the allstars negative control from qiagen + Hiperfect transfection reagent. Hiperfect alone does not
appear to be toxic...

I was under the impression that MCF7 was fairly tolerant to siRNA. Does anyone recognise this or have any theories?

Well.....
I would say that maybe your target gene is quite important for your cells!
Try with less siRNA (you could do a titration curve) and see what happens!!
