IHC on the liver: any special tricks needed? - (Aug/09/2008 )

Hi All,

I am currently trying to do IHC Ki67, IRS1 and Foxo1 on liver. I have a slide that contains multiple mouse organs and I get good staining in all tissues except liver and skeletal muscle, even though I know from western blots that the proteins are expressed.

In fact, I have tested multiple different antibodies on that particular slide and there is not one that gives me any staining in the liver.

Is there some trick for IHC in the liver that I am not aware of? I didn't perfuse the tissue but I don't think that's the issue.

Any help would be greatly appreciated.

first of all, I am just curious, so you have slide which have multiple different organs in one slide??
well, I never have that kind of slide, and I usually optimize each staining depending the organs.

anyway, liver sometimes hard to stain, because they are so fatty!!
but that does not nessesary mean you have to do something special.

so, my suggestion is (if you can) get slide which only have liver and optimize staining for liver.
if this didnt work, it's not a matter of staining, bacause Ki67 is at least not that hard.

in order to get better suggestion from better people (better than me)
tell us more about your staining, i.e. is that cryo-section/paraffin? and can you put your protocol??
