how to extract DNA from adenovirus for template of PCR - (Jul/31/2008 )

I have an adenovirus. I want to detect if the virus contained the fragment of my target protein by PCR

im not an expert on the subject but im gonna tell you just my experience and you can try something.
Ive worked in the past with lamda phage. to work with its DNA (the inserts) i had to isolate it from the phage with a kit.
at some point I realised that i could use the phage viruses as they were (without isolating the dna) in my PCR mix and they work just fine (i was amplyfing my target gene).
I dont say its the solution to your problem but is just a perspective. it worked for me...
