expression by hsp60 in E.coli - (Jul/25/2008 )

Hi ALL;

NOW am planning to make small exp. by using vectors with hsp60 promotor my question if I want to express the same vectors in E.coli what the methods to induce the expression by hsp60 in E.coli or M.Smegmatis and what the best methods for the purification if I want to use my protein in Animal model.

thanks in advance

Sorry, Longer. But I guess you have to wait a little more for somebody with experience in protein expression for animals, particularly for Inter-vascular use. (I am assuming you are going to inject the protein of interest into your mouse/rat), and experience with Mycobacterium smegmatis.

And correct me if I am wrong, but isn't hsp70 a Eukaryotic promoter. A hsc promoter would be the Prokaryote equivalent. In any case, I don't think heat shock promoter is the best thing to use in for protein expression for in E.coli. An example of a commonly used protein expression promoter is the T7lac promoter. A google search should pull up other promoter used for protein expression.

thank u perneseblue,

yea ur right, but the hsp60+hsp70 are presents in mycobacteria, rickettsia, coxiella and chlamydia

anyhow thanks again
