biotin-streptavidin hibridization - how can we detect biotin labeled RNAs with streptavidin HRP (Jul/13/2008 )

We want to detect biotin labeled RNAs that hybridized with their antisense (RPA reaction) by streptavidin HRP. After crosslinking of RNAs to BrightStar-Plus Nylon Membrane, we want to hybridize biotin and streptavidin. What buffer would you recommend as blocking and washing buffer while detection of biotins with streptavidin HRP for detection biotin labeled RNAs. And in which buffer should we incubate streptavidin with membrane.

Why do you go with the biotin-system? I would prefer the DIG-system (digoxigenin) which is easier and more sensitive (less background)....
