Fresh tissue slicing - need to fix in two different fixatives (Jul/10/2008 )

Does any one have some advice for slicing fresh mouse placenta (~1cm diameter). I have to slice into 4-6 slices, and sample these alternately into Glutaradehyde/Formaldehyde fixative (for resin-embedding) and Formalin (for paraffin wax embedding). Unfortunately due to my vertical orientation it is not possible to use a vibratome so I need to slice manually...

Any advice very gratefully received!

First fix the entire placenta, or the entire conceptus overnight. Then do histo-processing.

At the time of embedding (step prior to cutting the block), keep them in sideways position (placenta), or just let it lie as it would by itself (conceptus).

Histotechs should help you with keeping hte placenta sideways (on edge), there are many tricks.
