buffer preparation - (Jul/10/2008 )

i need to prepare a buffer,the composition is Tris ph7.5,x,x,x,x,. what should be the final pH of my buffer.should it be the same as 7.5 or if the use of Tris ph 7.5 would do?

let us know the name of buffer, and what would you like to do with that,

My question is common for preparing all the buffers.example lysate buffer says
15mM Hepes ph7.5
250mM Nacl
0.5%np-40
10%glycerol
25mM NaF.

If i am using Hepes ph7.5 from stock,will it do or i have to adjuse the PH of the final buffer to 7.5?

you want the final pH of your buffer to be 7.5.

we use a stock at the proper pH and usually don't need to adjust it (depends on additives). you can check and adjust prior to final volume adjustment if you feel it is necessary.
