How to culture Top10 with pBI121 ? - (Jul/08/2008 )

I am trying to culture Top10 cells containing pBI121 on LB Broth with 50 microgram/ml in Kanamycin but i get nothing. ANY IDEA ?

Most likely your cells lost the plasmid.
I would retransform and try to start a culture from a fresh colony
