RNA extraction from skin tissue - (Jul/07/2008 )

I want to know, how the good method to homogenize skin tissue? I have experience it is so difficult to do it in the initial step of RNA extraction. I use rotor, but the tissue undestruct. the protocol suggest nitrogen treatment to homogenize it, but unfortunately it so difficut to get it, and so easy to loss cause by evaporate. Any other manner?

Several ways:

1. Tissue grinder

2. Homogenizer

3. Liquid Nitrogen

4. Dismembrator

5. Ultrasonic homogenizer

6. Polytron (best)

Try to homogenise using liquid N2 and mortar and pestle. Should work fine (dont forget to pre-cut it).
