Primer extension and sequencing from total RNA - (Jul/03/2008 )

I would like to know if I can use the classical primer extesion to sequence an RNA using ddNTP + dNTP. Thanks.

You couldn't use total RNA, you'd get way too much false priming and the background would be too great. Even if you had just your RNA of interest, you might struggle to get enough RNA to do this. Sequencing requires much more template, it's not a PCR where you only require a small amount of template, rather it's a linear increase in product so you need a lot more to start with.
