Best Fluorescent Protein - (Jul/02/2008 )

Hello,

I'm trying to design fluorescent protein chimeras for monomeric GTPases.

my goal is primarily to do live cell imaging of these fusions in human smooth muscle cell cultures.

Which fluorescent protein is the best choice for this application?

blue? yellow? cyan? or just plain GFP?

I would probably go with eGFP. Time tested.

I've got an additional question - when making fluorescent protein (FP) fusions, do I need spacer amino acids included between the FP and my protein of interest? how many? which amino acids are best to use?
