DTT preparation - (Jun/24/2008 )

Hi all,
I was just wondering what kind of water should we use to dilute stock solution say like DTT, NaOH, SDS, NaAc,...etc. is it Distalled or nanoPure H2O. Do i need to autoclave the solutions before using them in tissue analysis (DNA extraction) or there is no need for that.

Another question is how much EtBr shouls i add for 50ml LE agarose gel?

About Etbr conc ,It can be used in a concentration range between 0.5 and 1 µg/ml. we use 50 µl of 0.5 mg/ml Ethidium bromide solution for 50 ml agarose solution. you can try it out and see if it works

we use milli-q (same as nanopure, different brand deionizer) water.

do not autoclave dtt, naoh or sds. if you need to sterilize any of these (and, i don't see why you would, we never do) then filter sterilize or prepare them with sterile water.
