X-Gluc storage - (Jun/18/2008 )

Hi

We bought X-gluc 2 months ago and it was storage at RT, but it was supposed to be storage at 4 C.... The person That did it didin't read the instruction I guess. angry.gif Now I need to use it so I prepare new buffer form it. The positive control is staining quite well, but the rest of the samples are not...I am supposed to see at least some staining at the tips of the leaves and root but it what i see is far weaker than what I am supposed to see....
Since the positive control is ok, I think the X-gluc can still be used (I put it at 4c now) but I am concerned about the other samples....Do you think it could be because of the X-gluc went bad?

Did you try a longer staining period, and then bleach the samples with ethanol? That might help. It could be that the x-gluc went bad, but then again it's hard to say if you've never stained these samples before. I guess I would try more positive controls before coming to that conclusion.
