Lysis buffer - (Jun/18/2008 )

Is it necessary to prepare stock of Tris-HCl and EDTA separately in preparing lysis buffer. What I have right now is TE buffer (50mM Tris-Hcl, 5mMEDTA, PH (8.0), is it possible to add it instead of the separate stocks. i mean is there any way to adjust the quantites to the ones required in preparing the lysis buffer.

The only problem I see is that you'll be using a 1X buffer to make up another buffer. If you are only adding a small amount of supplementary reagents (detergent, protease inhibitors, etc) then I guess it could be okay, but it won't work as well if you have to add more Tris or EDTA. I would just forget the already prepared TE, make up your own 2X stock of "lysis buffer salts" and use that.
