RNA gel loading buffer? - (Jun/17/2008 )

I have a gel loading buffer from the transcription kit. It said that the applications are "polyacrylamide urea gel electrophoresis" and "non-denaturing agarose gel electrophoresis. (http://www.ambion.com/techlib/spec/sp_8547.pdf)

I want to run a denaturing agarose gel, can I still use this loading buffer?

When do one want to run a non-denaturing RNA agarose gel?

I think when one just needs to check presence/absence and overall integrity of RNA in the sample.
Check the 1st ambion link, don't be deceived by the title: you will have to go at the end of document to find info about native gel:

no. you need denaturing loading buffer.

Ok. But the loading buffer contains mostly formamide, isn't that denaturing?

yes, formamide is denaturing. i hadn't read the sheet that you referenced prior to my response. you can use the loading buffer for denaturing gel samples, just follow the instructions.

in some denaturing loading buffers, formaldehyde is used instead of formamide.

Formamid definitely works better for denaturation. For 100µl of loading buffer I use 72µl formamide, 36µl Glycerol and some Bromophenol blue. I add 10µl of the buffer per µl of sample. It works really well.
