Reverse transcriptation for qPCR with some specific primers. Should I use primer - (Jun/13/2008 )

Hi all member

I would like to run RT-qPCR with specific primers for RT step. I have 2 primers, one for mRNA of interest and one for mRNA of reference.
Normally, in almost publication, author use random hexamer for priming in RT step, then they will get the mixture of cDNA including target cDNA and reference cDNA. Then the cDNA mixture is used as the template for qPCR

So, I would like to ask you: should I use 2 primers together in one RT reaction or single primer in each RT reaction?
In the case I use both primers in one RT reaction, I will get the mixture oF 2 cDNAs ( It is similar to the case use random hexamer).
If I use them seperatly, I will get two cDNA products.

I think you can put 2 primers together in one RT reaction, but make sure two pairs do not interfer with each other.

Actually, in case of using random hexamer, each primer have to compete with others.
