best way to extract HIV RNA - Trizol vs Qiagen (Jun/12/2008 )

Hi there!

I have noted that almost all the reported work dealing with plasma HIV RNA, extract the nucleic acid using Qiagen's columns. For me, that works great. However, conventional scientists still believe that Trizol will make it better. I have tried many times with Trizol (and its concetrated version) and it simply does not work.
I am wondering why, if you extract total RNA with trizol, then you should have more chances to catch the viral RNA!!

Any advice??

I may be wrong but people use Trizol for HIV RNA, so it should work; some people prefer subsequent qiagen column purification.

dear gradstudent

yes recently tried to isolate viral RNA from HIV samples using TRIzol.. It worked fine and i could isolate 30ng/ul... which is pretty good amount... although it was not highly pure but my real time PCR showed good results

can you send me a protocol you have been using... so that i can trace where you need to improve?

thanx
