loxP sites - (Jun/11/2008 )

Hi All,
How can i find loxP sites in a given sequence.
When I try to amplify the large fragment… I’m getting unexpected bands in the PCR.
If it is becoz of loxP sites… how can I overcome this problem?

I am not sure what you mean but.. You must have LoxP site sequence..

1. By sequence alignment
2. Sequencing.

If the fragment that you are amplifying has a loxP site in it, the PCR may give trouble (because of the inverse repeats of LoxP), but it works. I have done such PCRs. You just have to work on PCR conditions.
