RNAi effect-timing - (Jun/09/2008 )

Hi, how long after microinjection of dsRNA does it take until the RNAi effect happens? I mean, what is the shortest time, after which I can be sure, that the effect is due to RNAi and not microinjection. Thanks

It varies. In cell culture, I generally find it 48-72 hours for a maximum effect. In your case, and with your target RNA it may be different. THe only way to make sure (and to demonstrate to your critics) is to look for RNA and protein levels at various time points. If cell culture, northern, RPA, RT-PCR and western. If embryos, in addition to the others, in-situ hyb.
