Fluorescent Label Removal From Ab - (May/30/2008 )

Hello,

We are trying to remove the dye label (Texas red) from some Rabbit polyclonal Ab's, and was wondering if anyone had any idea on how to do this?

I know its probably an odd request, but we are trying to get rid of the Ab and analyse the dye presence using our own method.

We have tried to shock it off using 0.1M NaOH, but didnt recieve any signal.l

Any help on this would be greatly appreciated.

If you can assume the coupling is in the fc region why not digest the ab part with papain or pepsin? You will get your dye with a small portion of the ab still attached. One of the enz will digest above the S-S bond; one will digest below.

you will get F(ab')2 and also Fc, or F(ab)'

I dont see any easy ways to remove conjugated dyes from antibodies without killing antibody activities. The coupling position should be ramdom. Can you justify why would you need to do so?
