Na Acetate buffer recipe - Na Acetate buffer (May/15/2008 )

Hello Everyone,

I am a bit confused in preparing acetate buffer using na acetate and acetic acid. i need to prepare 10 mM na acetate buffer pH 4.0 and 25 mM Na Acetate buffer ph 4.5 and one with 25 mM Na Acetate buffer ph 4.5 containing 0.4 M Nacl. how much quantity of sodium acetate and acetic acid do i have to add to get the exact pH. can i prepare concentrated solutions and later dilute them as per the use.

According to a recipe book from Roche Biochemicals

For a 3M solution pH 5.2

408.24g of Sodium Acetate (3H20) in 800ml of DD H20 and adjust pH 5.2 with glacial acetic acid Make it 1 liter sterilize by autoclaving

since you are preparing a couple of different concentrations of buffer and one with added salt, your best bet would be to prepare a higher concentration stock at the two pHs that you need.

to prepare the stocks, i would make a 0.1M acetate buffer at each pH.

make a 0.1M stock of acetic acid and a 0.1M stock of na acetate.

mix them together until it reaches the target pH(s). by doing it this way, no matter what the final volume or pH it will still be 0.1M buffer.

then make your final dilutions from the stock (10x for 10mM and 4x for 25mM, and don't forget the salt).
