Help: Genomic DNA PCR - (May/12/2008 )

closedeyes.gif Hi everybody, I did genomic DNA PCR these days. While from same sample, some primers couldn't get result, some primers have nice result. So can you give me some reason for this phenomenon? Thanks closedeyes.gif

Look up some protocols here:

But in short: it depends upon the primer design, if some of your primers are not desgined well, or they have false annealing to some other genomic region you may not get specific products for these.

another possibility is that the annealing temperature is not the optimal, therefore no amplification.
