siRNA transfection in endothelial cells - (May/08/2008 )

I am trying to use endothelial cells as target for my siRNA.

It was known that it is harder to transfect plasmid into endothelial cells. are there any better methods for transfection siRNA into endothelial cells?

for example, lipofectamine 2000? lipofectamine LTX? FuGENE6? Oligofectamine? Nucleofection? shRNA virus? Any suggestions?

Thanks

ithink it depends on which cells are you using.
i tried fugene6 and siRNA transfection reagent on HUVEC and they both work well, given that you change medium after 5-7 hours. after 24 hours you have toxicity.
lipofectamine 2000 started to kill my cells (ANY kind of cell!) 4 years ago quite abruptly, i've never been able to use it anymore, not that i complain.
Think you have to try.

I appreciate your reply.

Do you have the protocols for transfection ratio of FuGENE6 with siRNA on HUVEC? May I ask for it?

Did you check the transfection rate after transfection?
