Stripping Solution: Recycle Anyone? - (May/08/2008 )

Dear all,

yesterday I prepared a stripping solution as follows:

beta-mercapto ethanol --> 560 uL
SDS --> 2%
0.5M tris pH 6.8 --> 10 mL
water up to 80 mL


My nitrocellulose membrane was then incubated for an hour at 60°C and then collected the used solution in a test tube.

Can I use this solution once again to strip the same membrane or would I get better results with a fresh stripping solution?

Hi,

I don`t know if it would help you but in our lab we always prepare fresh solution.

Fresh will be ideal and using mercaptho is pretty harsh on your blot. should try the milder one with glycine. It works fine too!

Sorry: I nearly forgot about this question. Well, given that no one replied glare.gif I decided my lab could afford some SDS and mercapto ethanol and prepared it fresh. Mercapto ethanol is harsh, indeed, but the result is always excellent (nitrocellulose really looks brand new).

By the way, if anyone has a (better) alternative to my stripping solution, please, feel free to share it: I am always willing to try new recipes. cool.gif
