FACS analysis - (Apr/22/2008 )

I'm going to perform FACS on my Huh7 cell lines for CD44 cells. I'd like to know do i trypsinise the cells in trypsin or just EDTA? thanks

Well if you want to analyse cell surface markers you should NOT use trypasin as it will destroy your proteins by cliving them of. EDTA is milder and work most of the times

Thank you for your suggestion. I'm new to FACS. Can you tell me what we mean by gauging the cells? How do we know what area to gauge?
