Can some one tell me the principle behind the determining the protein concentration. Which amino acids are target for the absorbance of UV and which wavelength are normally used. And how to see its purity??

you look at aromatic amino acids at a wavelength of 280nm.

you look at peptide bonds with a wavelength of ~215nm.

you determine purity on page.
