Beta mercapta ethanol - (Apr/09/2008 )

Why the Beta mercapta ethanol is used in the SDS gel

-julebo-

Beta mercapta ethanol is used to disrupt the disulfide bond.

but SDS is also a strong neturating agent, it does not break the Disulfide bridge,
in nondenatruing condition either B mercapta ethanol should be added or not.

SDS negatively charged the proteins and unfolded their tertiary structure, however, SDS can't disrupt the disulfide bridge as it is covalent bond.
In nondenaturing PAGE, 2-mercaptoethanol should not be added to the sample buffer.
