Does the nitrocellulose need to be pre-wetted (like in traditional western blot), or can you just spot the proteins directly onto the dry nitrocellulose?

If I am testing antibody dilutions for westerns, I dot cell lysates onto dry membrane and then wet the membrane before blocking. It works

it will not really soak in; better you use a dot blot apparatus where you need no pre-wetting
